You’re all set! Dave Call, Gregg’s general manager, said the company has invested about $1 million in changes to the facility to address nuisance odors and to comply with AQMD orders. “I have put forth a good-faith effort and I felt our efforts will be rewarded,” Call said. “I’m encouraged by the board’s decision. We will continue working on projects to improve the number of environmentally friendly materials here at Gregg.” Last April the AQMD issued an order of abatement, the hearing board in November issued 16 requirements that the foundry, located at 10460 Hickson St. near Rio Vista Elementary School, needed to comply with in cleaning up its operations. The requirements included switching to less odorous sand for the company’s molding operations and ceasing casting operations between 3 and 9p.m. Gregg is scheduled to meet with the AQMD hearing board again in July for a progress report. EL MONTE – The South Coast Air Quality Management District has decided to allow an iron foundry to stay in business. The AQMD began monitoring El Monte-based Gregg Industries last February after residents accused it of emitting foul odors into the air. After months of public hearings, the AQMD’s hearing board issued an amended order Tuesday that said odor levels have decreased, residential complaints dropped significantly, from 46 in January to 12 in February, and that the company passed an odor performance demonstration. AQMD officials added they do not expect the order to result in the closure of Gregg Industries. last_img read more

first_imgSome Cambridge scientists engineered a four-character genetic code and made some proteins with it.  They guided the process at every step, but claim that they “evolved” this code.  Is that a fair use of language?  This strange admixture of concepts is found in today’s issue of Nature.1  The confusion began right in the title: “Encoding multiple unnatural amino acids via evolution of a quadruplet-decoding ribosome.”…orthogonal pairs have been evolved to incorporate a range of unnatural amino acids…Here we synthetically evolve an orthogonal ribosome (ribo-Q1) that efficiently decodes a series of quadruplet codons….Because the synthetasetRNA pairs used have been evolved to incorporate numerous unnatural amino acids it will be possible to encode more than 200 unnatural amino acid combinations using this approach.….this work provides foundational technologies for the encoded synthesis and synthetic evolution of unnatural polymers in cells.Natural ribosomes are very inefficient at, and unevolvable for quadruplet decoding, which would enhance misreading of the proteome.…orthogonal ribosomes … may, in principle, be evolved to efficiently decode quadruplet codons on the orthogonal message….To discover evolved orthogonal ribosomes that enhance quadruplet decoding we first created 11 saturation mutagenesis libraries in the 16S ribosomal RNA of ribo-X (an orthogonal ribosome previously evolved for efficient amber codon decoding on an orthogonal message)….We used ribo-X as a starting point for library generation because we hoped to discover evolved orthogonal ribosomes that gain the ability to efficiently decode quadruplet codons while maintaining the ability to efficiently decode amber codons on the orthogonal mRNA….To explicitly compare the fidelity of triplet decoding and quadruplet decoding for the evolved orthogonal ribosomes and the progenitor ribosome we used two independent methods….….which is derived from the … pair, by a series of generally applicable directed evolution steps….…it will be interesting to investigate the enhancement of protein function that may be accessed by combining the encoding of these cross-links with directed evolution methods. What’s notable in this paper was not only the flagrant use of evolve as an active method that the scientists used to investigate function for the purpose of enhancing protein synthesis, but also their use of the stem word “natural.”  They spoke of unnatural amino acids (those not found in wild-type living cells), and described their attempts to achieve of “synthetic evolution of unnatural polymers in cells.”  They spoke of the “inefficiency with which natural ribosomes decode quadruplet codons” but then praised their fidelity with their triplet system: “Natural ribosomes decode triplet codons with high fidelity (error frequencies ranging from 10-2 to 10-4 errors per codon have been reported).”    Another mixing of unguided and designed concepts appears in their use of synthetic.  They spoke of “synthetic evolution” (their lab work) but also used and altered the “natural” molecular machines that bind amino acids to transfer-RNA, the tRNA synthetases.  Consider also how the concept of coding was used: they wished to achieve “synthetic genetic code expansion” They shifted seamlessly between the natural genetic code and their expanded, synthesized quadruplet code.  With all this mixture of terms synthesizing and evolving natural and unnatural codes, the reader is left wondering what “evolved” on its own, if anything, and what was intelligently designed.1.  Neuman, Wang, Davis, Garcia-Alai, Chin, “Encoding multiple unnatural amino acids via evolution of a quadruplet-decoding ribosome,” Nature 464, 441-444 (18 March 2010); doi:10.1038/nature08817.Answer: everything was intelligently designed, both the natural and unnatural codes and functions.  This paper was one of the best examples in recent memory of Truman’s Law: “If you can’t convince them, confuse them.”  Using evolve as a synonym for design is a clever way to blow smoke using equivocation.  Words mean things.  This has nothing to do with evolution in the way Darwin used it, and in the way the debate rages today.  It has everything to do with intelligently designing codes to synthesize things they would not naturally do (that is, without the intervention of a human mind).  These human designers did not “evolve” anything, and they did not rule out intelligent design in the “natural” systems.  If they really wanted to talk about evolution, they should have left the lab and let “nature” take its course.  “Synthetic evolution” is as sophoxymoronic (02/02/2008 commentary) as holy atheism.(Visited 31 times, 1 visits today)FacebookTwitterPinterestSave分享0last_img read more

first_imgShare Facebook Twitter Google + LinkedIn Pinterest The first positive case of West Nile Virus (WNV) in an Ohio horse has been confirmed in 2016.Testing on samples taken from a seven-year-old Standardbred in Tuscarawas County confirmed the positive WNV diagnosis to the Ohio Department of Agriculture Sept. 12. The horse’s veterinarian first examined the animal Aug. 29. The animal was euthanized after exhibiting significant clinical signs, including shaking, agitation and thrashing. The horse had not been vaccinated.West Nile Virus is transmitted to horses via bites from infected mosquitoes. Clinical signs for WNV include flulike symptoms, where the horse seems mildly anorexic and depressed. Changes in mentality, drowsiness, driving or pushing forward (often without control) and asymmetrical weakness may be observed. Mortality rate from WNV can be as high as 30% to 40% in horses. Infection with WNV does not always lead to signs of illness in people or animals. WNV is endemic in the United States and Ohio has reported three positive cases in horses each of the last few years.“This incident in Tuscarawas County should serve as an alert to all horse owners to vaccinate their animals against West Nile Virus,” said State Veterinarian Dr. Tony Forshey. “Vaccines are a proven and effective prevention tool and I encourage all owners to talk to their local vet for options and advice on how to keep their animals healthy.”In addition to vaccinations, horse owners also should work to reduce the mosquito population and eliminate possible breeding areas. Recommendations include: removing stagnant water sources; keeping animals inside during the bugs’ feeding times, which are typically early in the morning and evening; and using mosquito repellents.last_img read more

first_imgEvery once in a while, a piece of gear comes out that threatens to change everything. In this case, it’s ARRI’s new Orbiter.Nothing excites me more than seeing significant advancements in the lighting field. Cameras are constantly getting updates, but in the grand scheme of things, most cameras already look cinematic, offer good slow motion, and shoot in 4K or higher. To me, the lighting field is where truly cinematic images begin — and where the advancements are most apparent. Just look at the way the Quasar changed the lighting landscape, creating an entire style of music video lighting altogether. This week, ARRI announced the Orbiter. It’s appropriately named, because it looks like something straight out of the latest Sci-Fi movie. The Orbiter, to say the least, looks sick.In some regards, this light represents the first lighting unit that I would firmly say can do anything. It can be a leko light, it can be a softbox, it can be a fresnel, it can hook up to other lights via ethernet — and you can control it wirelessly via DMX on your iPad. It can automatically gauge the light in the room and change seamlessly to varying degrees of brightness and color temperature. It can do many things.Key Features:Variety of optics, including open face, projection, dome, and light banksARRI Spectra six-color, wide-gamut light engineExtremely powerful output for maximum brightness and perfect colorsLighting Operating System (LiOS) with powerful software featuresWeatherproof housingIntegrated color sensor for matching ambient lightRemovable, intuitive control panelFull suite of connectors and sensorsPerfected smooth dimming to zeroInternal power supply, wireless DMX, and battery inputOne thing that I initially thought about this release was how it almost seems like ARRI’s version of many the things that Aputure announced at NAB this last year. Also, the addition of the quick change optics system reminds me quite a lot of the 120 and 300d lighting units (though, this light doesn’t feature a bowens mount like the others, requiring proprietary ARRI modifiers). The Orbiter is obviously a higher-end model, which means a higher price-tag, but these features seem to mirror much of what Aputure is now and will soon be offering. The Orbiter definitely has quite a few impressive tricks up its sleeve. Here are the ones I’m most excited about.Quick Lighting MountARRI has developed a new Quick Lighting Mount system for the Orbiter. Unlike some other products on the market, this system does not involve a bowens mount; rather, it includes their new QLM (quick-lighting mount). This means that if you want to use various modifiers, you’ll need to purchase them from ARRI.So far, this system seems to involve an open face option, a projection lens option (similar to a source four leko), a dome option, and an adapter to allow for soft banks (similar to a chimera system).ARRI Spectra Light EngineARRI units have always impressed with the quality of the light they create. This unit represents a leap ahead of the competition in this regard. ARRI Spectra is a six-color light engine. According to the product pageIncluding a red, green, blue, amber, cyan, and lime LED, the ARRI Spectra six-color light engine translates into a wider color gamut, more accurate colors, and most importantly, higher color rendition across the entire CCT range. This results in an unprecedentedly high CRI of about 98. This engine also includes a color sensor mode, which will read the ambient color surrounding the fixture and match the color with complete accuracy. It can also accurately render colors from 2,000 to 20,000 kelvin. This is the future of lighting — LED lights with high output and nearly flawless color rendition.Fully Waterproof HousingWhile there are certainly more technical specs that are impressive about this light, this one for some reason really catches my attention. This light is fully waterproof. The fact that a light with this much technical capability, output, and everything else can also operate it in any scenario, including rain, is just incredible. This light just seems like the answer to any lighting situation.I think this light represents a shift (that I think other companies had already started) in the lighting industry. LED lighting has now shifted from being a competitor to more traditional lighting styles (HMI or Tungsten) and is now creating a class of lighting all its own. In 5 years or less, HMI or tungsten units may be a thing of the past on the majority of movie sets. We’ll see.Cover image via ARRI.Looking for more on film and video gear? first_img2 ‘newbie’ drug pushers fall in Lucena sting FILE – In this file photo dated Wednesday, Jan. 11, 2017, Athletic Bilbao’s Aymeric Laporte in action during Copa del Rey, 16 round, against FC Barcelona at the Camp Nou in Barcelona, Spain. 23-year-old Athletic Bilbao says defender Frenchman Aymeric Laporte is reported Monday Jan. 29, 2018, to have paid his buyout clause, looks set to leave the club, opening the way for a transfer to Manchester City. (AP Photo/Manu Fernandez, FILE)MANCHESTER, England — Manchester City signed France under-21 international Aymeric Laporte from Athletic Bilbao for a club-record fee of 65 million euros ($80 million) on Tuesday, taking its spending on center backs since 2014 to about $250 million. The 23-year-old Laporte came close to joining City in 2016 — when Pep Guardiola took charge of the current Premier League leaders — but backed out of the move after feeling the Frenchman wasn’t quite ready.ADVERTISEMENT Vincent Kompany, the club’s other center back, has been at City since 2008 and is the captain. But the Belgium international might be surplus to requirements from next season, especially considering his injury record.City also spent about 120 million pounds (then $155 million) on three fullbacks — Kyle Walker, Benjamin Mendy and Danilo — in the last transfer window.“Last summer we had a problem, with six or seven players out of contract and over 30 years old, so we needed to make a change,” Guardiola said. “We decided to sign four or five different players and at this stage, Manchester City, when you want to compete at the highest level, you need to spend.“One day, maybe this club will spend 130-200 million pounds on one player. So far, we haven’t had to do that.”Laporte made his debut in 2012 for Bilbao under Marcelo Bielsa — a coach revered by Guardiola — and quickly became one of the Spanish league’s most promising players. He said he likes to bring the ball out from the back, giving him the pedigree that suits Guardiola’s vision and making him a likely rival for Stones to play alongside Otamendi.ADVERTISEMENT Don’t miss out on the latest news and information. I also like playing long, cross-field passes to change the position of an attack and give us another option. Variety is important.”Barcelona has previously been linked with Laporte, who has similar qualities to Gerard Pique. The size of the fee reflects the shortage of top-quality, left-footed center backs around at the moment — and comes two weeks after City turned down the chance to sign Alexis Sanchez partly because of the Chile forward’s financial demands.Laporte has signed a contract until 2023 and becomes City’s most expensive player, with the fee exceeding the reported 55 million pounds spent on Kevin De Bruyne in August 2015.City has four of the five costliest defenders in Premier League history — Laporte, Stones, Walker, and Mendy.Laporte, who hasn’t played for France’s senior team, will be available for the knockout stage of the Champions League.“All I know,” he said in an Instagram message to Athletic’s fans on Monday, “is that I will never forget the place where I’ve become a better player and a better person.
